Summary and additional information:<- Kerstin Fiege and Nicole Frankenberg-Dinkel, Technical University Kaiserslautern; Nissle 1917 (T7); EcNT7. Germany. Strain carries chromosomal integration of T7 polymerase under control of lacUV5 promoter; use for the heterologous expression of heme cofactor; therefore, strain is suitable for expression of genes under control of T7 promoter; the strain is unique in that it is able to take up heme intrinsically from culture media so that heterologously produced proteins can be loaded better with heme cofactor. The plasmid used for gene integration of T7 polymerase into the genome (pUC-T7-FRT-Kan) was removed from chromosome by FLP-FRT-site technique, strain carries neither the plasmid nor antibiotic resistances, vector was excised from the genome by lambda recombination, strain does not contain any remains of the plasmid. The T7 polymerase is necessary for gene expression under control of the T7 promoter, used in expression vectors like the pET or pDuet.
Please note, very important! E. coli strain IPC-ntb2 mentioned in S. Ferrari et al., Int. J. Food Microbiol 388 (2023) and in ISO 10272:2017/AMD 2023 is NOT DSM 115365. IPC-ntb2 is DSM 116329!
