pSa143包含构巢曲菌的整个argB基因，去除含有酵母LEU2和2Jlm DNA的两个小EcoRI片段得到pSa123，向pSa123插入来自pKA5-1包含米曲霉补充蛋氨酸营养缺陷型突变的BamHI片段得到pRBMI。
