构建了aflR破坏载体pXHSE。pXN1中6.7kb含niad的HindIII-XbaI片段用T4 DNA聚合酶钝化,连接pACH,用SmaI和EcoRV预处理,再次钝端,去磷酸化生成pXHSE。pXHSE中的niad可选择标记取代了aflR编码区。
