从肺炎克雷伯菌基因组DNA中PCR扩增出puuC基因。将puuC基因克隆到pTAC和pLAC中,分别得到两个重组载体pTAC-puuC和pLAC-puuC。将载体转化到肺炎克雷伯菌中,产生了重组肺炎克雷克菌(pTAC-puuC)和肺炎克雷普菌(pLAC-puuC)。乳酸和乙酸合成基因ldh1、ldh2和pta的缺失遵循RecA同源重组的方案。通过菌落PCR和测序确认重组体。
