将含有pct基因的NheI-SpeI片段插入pduP序列下游的相应限制性位点(pBHA-pduP)。pBHA-pduP-pct-yahK是通过将含有pct(pBHA-pct)和yahK(pBHA-yahK)的NheI-SpeI片段顺序插入pBHA-pdulP的SpeI位点而产生的。pGEM-Placz-pduP-pct-yahK是通过将NheI-SpeI片段(含有来自pBHA-pduP-pct-yahK的pduP-put-yahK)插入pGEM-PracZ的SpeI位点而获得的。最后,用ScaI切割pBR322,用碱性磷酸酶处理,与用NotI消化pGEM-PlacZ-pduP-pct-yahK获得的DNA片段连接,然后用Klenow片段处理,得到质粒pBR-1,2PDO(pduP-pc-yahK)。
