| Comments： | BglII digestion of recombinant plasmid gives fragments of 5.2, 2.2, and 1.9 kb. Restriction digests of the clone give the following sizes (kb): EcoRI--5.4, 4.4; BamHI--9.8; PstI--6.2, 3.7; PvuI--9.8; BamHI/PstI--3.7, 3.4, 2.5. The depositor indicates that the BglII fragment sizes are slightly overestimated in this reference. EcoRI detects numerous bands, 3 of which are Y-specific. Male-specific band A (5 kb) is the cognate one and may correspond to 2 to 3 copies; maps to Yq11 and Yq12. Band B appears as 4.5 or 3 kb; maps to Yq11. Band C is about 1 kb and maps to Yq11. TaqI detects a 10 kb band equivalent to EcoRI band B (4.5 kb allele), an 8.5 kb band equivalent to EcoRI band A and B (3.0 kb allele), and a 3.8 kb band equivalent to EcoRI fragment A. The BglII subfragment of 2.2 kb preferentially detects the Y-specific bands. Use stringent wash conditions (0.1X SSC, 68C). |
