Restriction digests of the clone give the following sizes (kb): EcoRI/HindIII--2.686, 0.911; EcoRI/HindIII/SstI--2.686, 0.505, 0.406.
Sonicated extracts of E. coli HB101 transformed with this construct express both ST and LTB immunoreactivity.
The 0.406 kb EcoRI/SstI fragment encodes the heat stable enterotoxin, and the 0.505 kb SstI/HindIII fragment encodes the heat labile enterotoxin.
The latter fragment, derived from EWD299 (ATCC 37218), includes the coding region for the B subunit and 3 transcription and translation stop signals, but lacks the ribosome binding site and sequences encoding the signal peptide.
The former fragment is at the 5 end and includes the complete coding sequence and 5 regulatory regions (transcription initiation and ribosome binding site) but not the 3 transcription and translation stop signals.
