Absence of a transcription terminator 3 to the insert may cause instability of the plasmid after induction, due to interference with plasmid replication. When induced at 42C, interferon gamma (insoluble) can be produced to a level up to 14.7 percent of the total cellular protein. Consists of pPLc28 with the lambda PL promoter, a manipulated ribosome binding site from the E. coli trp attenuator, an engineered ATGTGTTAC sequence, and a sequence corresponding to nt 156-1170 of the sequence record. |
