Restriction digests of the clone give the following sizes (kb): PstI--5.8, 2.3, 1.9, 1.2; BamHI--8.2, 3.1; EcoRI--6.2, 5.4; BamHI/EcoRI--4.0, 3.5, 2.8, 0.5; PvuII/AvaI--4.5, 2.8, 2.2, 1.7; HindIII--4.5, 2.6 doublet, 1.9. Mucoid colony morphology on agar supplemented with ampicillin (0.5 mg/ml). The entire streptokinase (skc) structural gene is on the 2.5 kb PstI fragment. Clone expresses streptokinase of Streptococcus equisimilis which is exported into the medium. |
