Features (with orientation and position when available):
marker(s): kanR, ->
insert detection: lacZ, MCS: HindIII...EcoRI, ->
replicon: pSC101
Cross references:
Vector: pHSG-cre (plasmid)
Promoters: Promoter lac
Construction: pHSG298, lambdaKC
Marker(s):kanR
Construct size (kb): 4.0
Features: marker(s): kanR
other: cre
promoter: lac
replicon: pMB1
Applications:
produces protein recombinase, site-specific
vector for other uses
Comments:
Restriction digests of the clone give the following sizes (kb): EcoRI/SalI--2.6, 1.4; EcoRI--4.0; PstI--2.6, 1.4.
The cre gene contains the following restriction sites (approximate kb from the 5 end): BamHI--0.84; ClaI--1.29.
The lambda/plasmid hybrid vectors can then be used for automatic subcloning; that is, the cre-mediated excision of plasmid constructs.
A plasmid useful for generating novel lambda/plasmid hybrid vectors by using cre-lox recombination to exchange new plasmids for plasmids already in lambda/plasmid hybrids.
Constructed by ligating a 1.3 kb cre-containing fragment from lambdaKC to pHSG298 digested with EcoRI+SalI.
Media: ATCC® Medium 1236: LB Medium (ATCC medium 1065) with 25 mcg/ml kanamycin
Growth Conditions:
Temperature: 37.0°C
