IMPORTANT: To prevent amplification of a rearranged and/or deleted cosmid, we recommend streaking on LB + amp plates at 30C and picking small colonies for liquid culture. Probe used was a 7.6 kb TaqI fragment. Probe must be prehybridized with excess human DNA to compete out repeated sequences. Loci A, C, and D are in strong linkage disequilibrium. 29/40 (72%) individuals show heterozygosity at 1 or more loci. EcoRI detects an insertion-deletion polymorphism also detected by BamHI and BstEII. |
