An EcoRI/XhoI double digest excises the insert in 2 fragments of about 2.0 kb and 1.8 kb. There is an EcoRI site in the insert.
Restriction digests of the clone give the following sizes (kb): EcoRI--7.6, 2.1; HindIII--8.8, 0.9.
The coding region of this sequence differs from pcCSF-17 (ATCC 53149) by the insertion of 894 base pairs between nucleotides 721 and 722 of the latter (preserving the reading frame). The sequences diverge after the termination codon.
This sequence contains an open reading frame of 1662 nucleotides encoding 554 amino acids, as well as 2200 nucleotides 3 non-coding sequence.
The vector pXM is a mammalian cell expression vector. When this plasmid is introduced into COS-1 cells, it directs high-level expression of biologically active CSF-1.
