Disease: grade IV, adenocarcinoma
Age: 53 years
Gender: male
Ethnicity: Caucasian
Applications:
This cell line may be used as a transfection host.
Storage Conditions: liquid nitrogen vapor phase
Clinical Data:
53 years
Caucasian
male
Antigen Expression: HLA A23, A32, B15, B35; blood type O; Rh+
Genes Expressed:
carcinoembryonic antigen (CEA) 523 ng/10 6 cells/10 days; keratin
Cellular Products:
carcinoembryonic antigen (CEA) 523 ng/10 exp6 cells/10 days; keratin
Tumorigenic: Yes
Effects:
Yes, in nude mice
Comments:
The cells are negative for expression of CSAp (CSAp-) and colon antigen 3, negative.
There is a C -> T mutation in codon 248 of the p53 gene resulting in an Arg -> Trp substitution.

The cells are positive for keratin by immunoperoxidase staining.

Complete Growth Medium: The base medium for this cell line is ATCC-formulated Leibovitzs L-15 Medium, Catalog No. 30-2008. To make the complete growth medium, add the following components to the base medium: fetal bovine serum to a final concentration of 10%.

(Note: The L-15 medium formulation was devised for use in a free gas exchange with atmospheric air. A CO2 and air mixture is detrimental to cells when using this medium for cultivation)

Subculturing:

Remove medium, add fresh 0.25% trypsin, 0.02% EDTA for 10 to 20 minutes at 37°C. Add fresh medium, disperse cells and centrifuge to pellet the cells. Resuspend in fresh medium and dispense into new flasks.

Subcultivation Ratio: A subcultivation ratio of 1:2 to 1:5 is recommended

Medium Renewal: 1 to 2 times per week

Cryopreservation:
Freeze medium: complete growth medium, 95%; DMSO, 5%
Culture Conditions:
Atmosphere: air, 100%
