Strain: NIH/Swiss
Applications:
produces protein beta-galactosidase beta-D-galactosidase
Disclosure: This material is cited in a US or other Patent and may not be used to infringe the claims. Depending on the wishes of the Depositor, ATCC may be required to inform the Patent Depositor of the party to which the material was furnished. This material may not have been produced or characterized by ATCC.
Derivation:
The psi 2 BAG alpha cell line has produced an infectious helper virus upon prolonged passage, but present stocks do not.
The cell line was derived from the psi 2 cell line developed by Mann, et al. (Cell 33:153-159, 1983) from the NIH/3T3 cell line (ATCC CRL-1658).
Genes Expressed:
a beta galactosidase transducing vector (BAG),beta-galactosidase(beta-D-galactosidase) lacZ,beta-galactosidase
Cellular Products:
a beta galactosidase transducing vector (BAG)
Comments:
The psi 2 BAG alpha cell line has produced an infectious helper virus upon prolonged passage, but present stocks do not.
The BAG construct has been used to transfect psiCRE cells in ATCC CRL 1858 and has been used to transfect psi2 cells in ATCC CRL 9560.
This line produces a vector (BAG) that can infect and mouse and rat and transduce the bacterial beta galactosidase gene.
The BAG vector has been used to mark cells for lineage analysis based on the histochemical detection of the bacterial beta galactosidase.
The cell line was derived from the psi 2 cell line developed by Mann, et al. (Cell 33:153-159, 1983) from the NIH/3T3 cell line (ATCC CRL-1658).
This line may produce an infectious helper virus upon prolonged passage, present stocks do not.
A protocol to test for the helper virus is available and accompanies the cells when ordered.
The vector has been used to infect a variety of neural cell types and to express beta-galactosidase and resistance to G418.
Complete Growth Medium: Dulbeccos modified Eagles medium with 4.5 g/L glucose, 90%; bovine calf serum, 10%
Subculturing:
Medium Renewal: Every 2 to 3 days
Remove medium, add fresh 0.25% Trypsin - 0.02% EDTA and allow the flask to sit at room temperature (or 37C) until the cells detach (2 to 3 minutes). Add fresh medium, aspirate and dispense into new flasks.
Culture Conditions:
Temperature: 37.0°C
Name of Depositor: Harvard Medical School
U.S. Patent Number:
Passage History:
The psi 2 BAG alpha cell line has produced an infectious helper virus upon prolonged passage, but present stocks do not.
This line may produce an infectious helper virus upon prolonged passage, present stocks do not.
