Karyotype: hyperhexaploid; modal number = 142; range = 130 to 153. Each cell had over 70 marker chromosomes. Among the markers were t(1p10q), del(1)(q11/q12), 6q+, 2q+,der(20)t(12;20)(q11;q13). Some of these had 2 to copies per cell. Most cells also had 5 to 10 microchromosomes (metacentric like). Structurally normal N2,N4 and N9 were absent; two copies of the X and multiple copies of the Y were detected.

Comments:
The line lacks ultrastructural and biochemical evidence of squamous differentiation or mucin production.
The cells express easily detectable p53 mRNA at levels comparable to normal lung tissue, and exhibit no gross structural DNA abnormalities.
The cells stain positively for keratin and vimentin but are negative for neurofilament triplet protein.
Complete Growth Medium: The base medium for this cell line is ATCC-formulated RPMI-1640 Medium, ATCC 30-2001. To make the complete growth medium, add the following components to the base medium: fetal bovine serum (ATCC 30-2020) to a final concentration of 10%.
Subculturing:
Remove medium, add fresh 0.25% trypsin, 0.53 mM EDTA, rinse quickly and remove trypsin leaving 1 to 2 mL. Let the culture sit at room temperature (or at 37°C) until the cells detach. Add fresh medium, disperse cells and transfer to a new flask.
Subcultivation Ratio: A subcultivation ratio of 1:3 to 1:5 is recommended
Medium Renewal: 2 to 3 times per week

Freeze medium: Culture medium, 95%; DMSO, 5%
