The MH-S cell line was derived by SV40 transformation of an adherent cell enriched population of mouse alveolar macrophages.

Comments: The cells retain many of the properties of alveolar macrophages including typical macrophage morphology.
They are adherent, phagocytic, esterase positive and peroxidase negative.
Lipopolysaccharide (LPS) treatment stimulates IL-1 production.
The cells are capable of suppressing the in vitro plaque forming cell (PFC) response in a cell dose dependent manner.
Complete Growth Medium: The base medium for this cell line is ATCC-formulated RPMI-1640 Medium, Catalog No. 30-2001. To make the complete growth medium, add the following components to the base medium: 2-mercaptoethanol to a final concentration of 0.05 mM; fetal bovine serum to a final concentration of 10%.
Subculturing:
Cultures can be maintained by transferring floating cells to a centrifuge tube Rinse adherent cells with 0.25% trypsin, 0.53 mM EDTA solution. Remove the solution and add an additional 1 to 2 mL of trypsin-EDTA solution. Allow the flask to sit at room temperature (or at 37°C) until the cells detach. Add to floating cells collected above and centrifuge the cell suspension at 1000 rpm for 10 minutes, resuspend the pellet in fresh medium, aspirate and dispense into new flasks.
Subcultivation Ratio: A subcultivation ratio of 1:2 to 1:6 is recommended
Medium Renewal: Every 2 to 3 days
