Derivation:

The HT-2 cell line was derived in 1979 by J. Watson at the University of Auckland from a culture of C57BL spleen cells activated by sheep erythrocytes in the presence of IL-2.

This cell line was originally deposited as derived from the BALB/c strain of mice. Recent testing at ATCC and by the NIST Mouse STR Consortium using mouse STR technology has shown that the STR profile has more alleles in common with a C57BL mouse strain rather than BALB/c.

Comments:
The A5E clone was selected to provide a homogeneous population that is highly sensitive to IL-2.

In addition to IL-2, substantial proliferation is seen in the presence of interleukin 4 (interleukin-4, IL-4, also known as BSF-1).


Complete Growth Medium: RPMI 1640 medium with 2 mM L-glutamine adjusted to contain 1.5 g/L sodium bicarbonate, 4.5 g/L glucose, 10 mM HEPES and 1.0 mM sodium pyruvate and supplemented with 0.05 mM 2-mercaptoethanol, 100-200 IU/ml IL-2, 90%; fetal bovine serum, 10% Alternatively, IL-2 in the form of Rat T-STIM factor with Con A (available from Becton Dickinson, Cat.# 354115) may be used at 10% by volume.
Subculturing:
Cultures can be maintained by addition or replacement of fresh medium. Subcuture every two days at 3.5 x 10 4 viable cells/mL.
Medium Renewal: Every 2 to 3 days

Freeze medium: complete growth medium, 95%; DMSO, 5%
