Karyotype: This is a hypertriploid human cell line with the modal chromosome number of 80, occurring in 30% of cells and polyploidy at 4.1%. Most cells had a chromosome number ranging between 79 and 85. Seven marker chromosomes were common to all cells, including der(14)t(8;14) (q22;q24) (two copies), i(5p), der(6)t(3;6) (p21.1;p21.1), der(11)t(7;11) (q11.23;q23.1) and two others. Four other marker chromosomes occurred only once among 11 metaphases analyzed. Most cells had four normal X chromosomes. No Y chromosomes were found in QM preparation. Some cells had five copies of N1 and/or N3.

The established EB2 line is similar, in general, to EB1 but differs in that it is composed of larger and less uniform cells which grow more rapidly than EB1. Furthermore, the EB2 cells regularly form clumps, have more cytoplasm, a less rounded nucleus and microvilli. Electron microscopic studies reveal viral particles (Epstein-Barr virus), lipid bodies, vacuoles, poorly developed mitochondria, frequent microvilli and an unusual layered structure projecting from the nucleus (a finding also reported for EB1 cells). The line is considered more primitive in differentiation than EB1.
Complete Growth Medium: The base medium for this cell line is ATCC-formulated RPMI-1640 Medium, Catalog No. 30-2001. To make the complete growth medium, add the following components to the base medium: fetal bovine serum to a final concentration of 10%.
Subculturing:
Cultures can be maintained by addition or replacement of fresh medium. Establish new cultures at 4 X 105 cells/mL and maintain at between 4 X 105 and 1 X 106 cells/mL.
Medium Renewal: Every 2 to 3 days
Cryopreservation:
Freeze medium: Culture medium, 90%; DMSO, 10%
