DT95
规格:
货期:
编号:TS212245
品牌:Testobio
产品名称: DT95
商品货号: TS212245
Organism: Gallus gallus, chicken
Tissue: bursa
Product Format: frozen
Morphology: lymphoblast
Biosafety Level: 1

Biosafety classification is based on U.S. Public Health Service Guidelines, it is the responsibility of the customer to ensure that their facilities comply with biosafety regulations for their own country.

Disease: lymphoma
Applications:
DT95 is an avian leukosis virus (ALV) induced bursal lymphoma cell line derived from a Hyline SC chicken.
After one transfer in vivo, the DT95 cell line was established.
The cell line contains proviral DNA sequences integrated upstream from the c-myc proto-oncogene and expresses increased levels of c-myc RNA.
The cell line exhibits a more differentiated plasmacytoid phenotype than DT40 (see ATCC CRL-2111).
Infectivity assays revealed that DT95 does not release infectious RAV-1.
Storage Conditions: liquid nitrogen vapor phase
Derivation:
DT95 is an avian leukosis virus (ALV) induced bursal lymphoma cell line derived from a Hyline SC chicken.
Cell suspensions prepared from tumors that developed within the bursa of Fabricus were transferred intravenously into young syngeneic recipient chickens.
Oncogene: c-myc +
Genes Expressed:
immunoglobulin; IgM (surface and secreted),c-myc +,The expression of MHC class II is induced more rapidly by v-rel than c-rel and several weeks after infection, rel induced class II antigen as much as 50 fold more efficiently than c-rel.
Cellular Products:
immunoglobulin; IgM (surface and secreted)
Tumorigenic: Yes
Effects:
Yes, in 2 day old syngeneic chickens; tumors were observed in the spleens, kidneys and livers of recipient animals
Comments:
DT95 is an avian leukosis virus (ALV) induced bursal lymphoma cell line derived from a Hyline SC chicken.
The original lymphoma was induced by viral infection of a 1 day old chicken with Rous associated virus 1 (RAV-1).
Cell suspensions prepared from tumors that developed within the bursa of Fabricus were transferred intravenously into young syngeneic recipient chickens.
After one transfer in vivo, the DT95 cell line was established.
The cell line contains proviral DNA sequences integrated upstream from the c-myc proto-oncogene and expresses increased levels of c-myc RNA.
In vitro, both the c-rel gene and the v-rel oncogenes induce major histocompatibility (MHC) class I, MHC class II, and interleukin-2 receptor (IL-2R) antigens on the DT95 cell line.
The expression of MHC class II is induced more rapidly by v-rel than c-rel and several weeks after infection, rel induced class II antigen as much as 50 fold more efficiently than c-rel.
The cell line exhibits a more differentiated plasmacytoid phenotype than DT40 (see ATCC CRL-2111).
Infectivity assays revealed that DT95 does not release infectious RAV-1.
Complete Growth Medium: Dulbeccos modified Eagles medium with 4 mM L-glutamine modified to contain 4.5 g/L glucose, 1.5 g/L sodium bicarbonate and 0.05 mM 2-mercaptoethanol, 75%; tryptose phosphate broth, 10%; fetal bovine serum, 10%; chicken serum, 5%
Subculturing: Cultures can be maintained by addition or replacement of fresh medium. Start new cultures at 2 to 3 x 105 viable cells/mL and subculture before the cell density reaches 2 x 106 cells/mL.
Medium Renewal: Two to three times weekly
Cryopreservation:

Complete growth medium described above supplemented with 5% (v/v) DMSO.xa0 Cell culture tested DMSO is available as ATCC Catalog No. 4-X.

Culture Conditions:
Temperature: 37°C
Atmosphere: Air, 95%; Carbon dioxide (CO2), 5%
Name of Depositor: EH Humphries
Deposited As: Gallus gallus
