The CATH.a cell line was established from cultures of a tumor that arose in the brain of a transgenic mouse.

Comments: The transgene contain the SV40 T antigen coding sequence linked to a 783 bp portion of the rat tyrosine hydroxylase gene.
The cells express tyrosine hydroxylase and dopamine beta-hydroxylase, and contain neurofilaments.
Complete Growth Medium:

The base medium for this cell line is RPMI-1640 Medium (ATCC 30-2001). To make the complete medium add the following components to the base medium (88% final concentration):xa0xa0

  • Horse Serum (ATCC 30-2040), for an 8% final concentration
  • Fetal Bovine Serum (FBS; ATCC 30-2020), for a 4% final concentration
Subculturing:
Cultures are a mixture of adherent and suspended cells. Collect the floating cells and transfer to new flasks. Dissociate the attached cells using 0.12% trypsin without EDTA. Remove Trypsin by neutralization and gentle centrifugation followed by resuspension in complete growth medium and add to the suspension cell population.
Subcultivation Ratio: A subcultivation ratio of 1:4 is recommended

Freeze medium: 85% RPMI-1640 (ATCC 30-2001) + 10% FBS (ATCC 30-2020) + 5% DMSO (ATCC 4-X)
