Cell Type: hybridoma: B lymphocyte
Product Format: frozen
Morphology: lymphoblast
Culture Properties: suspension
Biosafety Level: 1

Biosafety classification is based on U.S. Public Health Service Guidelines, it is the responsibility of the customer to ensure that their facilities comply with biosafety regulations for their own country.

Storage Conditions: liquid nitrogen vapor phase
Disclosure: This material is cited in a US or other Patent and may not be used to infringe the claims. Depending on the wishes of the Depositor, ATCC may be required to inform the Patent Depositor of the party to which the material was furnished. This material may not have been produced or characterized by ATCC.
Derivation:

Animals were immunized with human lymphocytes.

Spleen cells were fused with NS-1 myeloma cells.

Genes Expressed:
immunoglobulin; monoclonal antibody; against CD45 (T-200)
Cellular Products:
immunoglobulin; monoclonal antibody; against CD45 (T-200)
Tumorigenic: Yes
Effects: Yes, forms ascites in pristane primed BALB/c mice
Comments:

The antibody has conventional CD45 reactivity and is included as antibody 815 in the panel used in the Third International Workshop and Conference on Human Leucocyte Differentiation Antigens.

The antibody has a higher affinity for CD45 than the BC8 antibody (see ATCC HB-10507).

Complete Growth Medium: The base medium for this cell line is ATCC-formulated RPMI-1640 Medium, Catalog No. 30-2001. To make the complete growth medium, add the following components to the base medium:
  • fetal bovine serum to a final concentration of 10%
  • 0.05 mM 2-mercaptoethanol

Subculturing:

Cultures can be maintained by addition of fresh medium. Alternatively, cultures can be established by centrifugation with subsequent resuspension at 2 x 105xa0viable cells/mL. Maintain cultures at a cell concentration between 1 x 105xa0and 1 x 106xa0cells/mL.

Medium Renewal: Add fresh medium every 2 to 3 days (depending on cell density).
Cryopreservation:
Complete growth medium supplemented with 5% (v/v) DMSO.xa0Cell culture tested DMSO is available as ATCC Catalog No. 4-X.

Culture Conditions:
Temperature: 37°C
Atmosphere: Air, 95%; Carbon dioxide (CO2), 5%
Isotype: mouse IgG2a
Name of Depositor: Fred Hutchinson Cancer Res. Cntr.
Deposited As: mouse (B cell); mouse (myeloma)
U.S. Patent Number:
