Comments:
Animals were immunized with metastatic human epidermoid carcinoma (HEp-3) cells by subtractive immunization.
Spleen cells were fused with NS0 myeloma cells.

A culture of 50-6 was deposited with the ATCC as PTA-227 in June of 1999. The cell line grown at ATCC is available as CRL-2696. The original patent deposit is available as PTA-227.
Complete Growth Medium: The base medium for this cell line is ATCC-formulated Dulbeccos Modified Eagles Medium, Catalog No. 30-2002. To make the complete growth medium, add the following components to the base medium:
  • O.1 mM Non-Essential Amino Acids (NEAA)
  • fetal bovine serum to a final concentration of 10%

Subculturing:
Protocol: Cultures can be maintained by the addition of fresh medium or replacement of medium. Alternatively, cultures can be established by centrifugation with subsequent resuspension at 1 to 2 X 10 exp5 viable cells/ml. Maintain cell density between 1 X 10 exp5 and 1 X 10 exp6 viable cells/ml.
Medium Renewal: Add fresh medium every 2 to 3 days (depending on cell density)
Cryopreservation:
Freeze medium: Complete growth medium, 95%; DMSO, 5%
Storage temperature: liquid nitrogen vapor phaseliquid nitrogen vapor phase
