Isolation:
Feces of dog with hemorrhagic enteritis
Product Format: frozen 1 mL per vial
Storage Conditions: -70°C or colder
Comments:
Non-pathogenic for neonatal pups. Antigenically indistinguishable from Feline panleukopenia virus and Mink enteritis virus, but has broader host range and different HA spectrum, especially in regards to pH conditions. Hemagglutinates porcine, Rh+ monkey, and feline red blood cells at pH 6.0-7.5. May fail to immunize dogs if passaged 8-10 times in feline cells.
Effect on Host:
CPE, refractile cells, cell rounding and sloughing
Recommended Host:
A-72 (ATCC® CRL-1542™)
Growth Conditions:
Recommendations for Infection: Plate cells 2-4 hours prior to infection and infect when cultures are 30-40% confluent. Remove medium and inoculate with a small volume of virus (e.g. 1 mL per 25 cm2) diluted to provide an optimal MOI (e.g. 0.1). Adsorb 1-2 hours at 35°C in a humidified atmosphere, rocking every 15 minutes to redistribute inoculum. End adsorption by adding virus growth medium.
Incubation: Incubate infected culture for 3-9 days at 35°C in a humidified atmosphere, until CPE are well advance throughout.
Virus Growth Medium: Leibovitzs L-15 Medium (ATCC 30-2008) + 2% FBS (ATCC 30-2020)
