Product Format: frozen
Storage Conditions: -70°C or colder
Disclosure: This material is cited in a US or other Patent and may not be used to infringe the claims. Comments:
Genome contains 18-nucleotide deletion between VPI nucleotides 3383-2301. Attenuated for chimpanzees at 200 ng dose.
Effect on Host:
Gradual deterioration of cells; titer by ELISA or by RT-PCR (cells become persistently infected). Subtle CPE may be missed. Monolayer does not fill in, cells round and deteriorate, but remain attached indicating persistent infection.
Recommended Host:
FRhK-4 (ATCC® CRL-1688™)
Growth Conditions: Temperature: 37°C
Recommendations for Infection: For best results, infection should be performed on a 70-80% confluent, 18-48 hour old cellular monolayer. Prepare dilution of virus in minimum amount of volume (e.g. 1 mL per 25 cm2). Wash monolayer with PBS or serum free medium prior to inoculation. Adsorb virus dilution for 1-2 hours at 37°C in a humidified 5% CO2 atmosphere, rocking every 20-30 minutes to redistribute inoculum. End adsorption by adding virus growth medium.
Incubation: 14-21 days at 37°C in a humidified 5% CO2 atmosphere.
