PA21 is one of the better characterized primate HAV strains that differs significantly from the HM175-like isolates Brown et al., J, Virol. 63: 4932-37, 1989. Hepatitis A antigen can be detected in infected cells by IF and in infected cell culture supernate by ELISA. This item has fungal contamination that can, with difficulty, be controlled with Amphotericin B.
Effect on Host:
No CPE, detect by IF, ELISA or PCR
Growth Medium: Minimum essential medium (Eagle) with 2 mM L-glutamine and Earles BSS adjusted to contain 1.5 g/L sodium bicarbonate, 0.1 mM non-essential amino acids, and 1.0 mM sodium pyruvate supplemented with 2.5 ug/ml Amphotericin B Solution and 2% fetal bovine serum
Recommended Host:
BSC-1 (ATCC® CCL-26™)
Growth Conditions:

Temperature:xa035°C

Incubation: 20 to 28 days at 35°C, a 5% CO2 in air atmosphere is recommended.

Recommendations for Infection: For best results cells should be 24 to 48 hours old and 70% - 80% confluent (not 100% confluent).
