Depends upon helper virus Human adenovirus 2 ATCC® VR-846™ for replication.xa0 Helper virus may be inactivated at 56°C for 30 minutes.

Resistant to heat 56°C 4 hours; this property can be used to selectively inactivate the helper adenovirus. Resistant to ether and chloroform.

Cross-reacts with AAV-3; no hemagglutinin detected. Depends upon helper adenovirus for replication.
Effect on Host:
Adenovirus CPE, refractile rounding and cell sloughing.xa0 AAV shows no CPE.

HEK-293 cells (ATCC® CRL-1573™)
Growth Conditions:

Temperature: 37°C

Recommendations for Infection: Plate cells 18-24 hours prior to infection and infect when cultures are 80-90% confluent. Remove medium and inoculate with a small volume of virus and helper virus(e.g. 1 mL per 25 cm2) diluted to provide an optimal MOI (e.g. 1.0 each for virus and helper virus). Adsorb 1-2 hours at 37°C in a humidified 5% CO2 atmosphere. End adsorption by adding virus growth medium.

Incubation: 2-3 days at 37°C in a humidified 5% CO2 atmosphere, until helper virus CPE is progressed through 80-90% of the monolayer. AAV does not cause CPE.
