Vaccinia virus ts mutant
Vaccinia virus ts mutant
规格:
货期:
编号:TS213562
品牌:Testobio
产品名称: Vaccinia virus ts mutant
商品货号: TS213562
Classification: Poxviridae, Orthopoxvirus
Agent: Vaccinia virus ts mutant
Strain: IHD-W Dts86
Applications:
The mutation was assigned by complementation analysis (Lackner, et al, 2003) to vaccinia map location A24, the 132-kDa RNA polymerase subunit.
Vaccination is recommended for laboratory staff manipulating vaccinia, monkeypox, and cowpox viruses.
Biosafety Level: 2

Biosafety classification is based on U.S. Public Health Service Guidelines, it is the responsibility of the customer to ensure that their facilities comply with biosafety regulations for their own country.

Isolation:
This conditional lethal mutant, originally designated plaque isolate number 9179, was isolated from the IHD-W strain of Vaccinia by S. Dales. The IHD-W strain was derived from an IHD-J strain. This mutant was selected after mutagenesis with nitrosoguanidine and replication in the presence of bromodeoxyuridine. This mutant was not assigned to a Dales EM category. The mutation was assigned by complementation analysis (Lackner, et al, 2003) to vaccinia map location A24, the 132-kDa RNA polymerase subunit. The non-permissive incubation temperature is 39.5°C.
Product Format: frozen
Storage Conditions: -70°C or colder

Effect on Host:
CPE: enlargement, plaques with rounding and eventual sloughing
Cytopathic effects in culture include enlargement and rounding of cells, followed by death and sloughing.
Growth Medium: ATCC® Medium 30-2002: Dulbeccos Modified Eagles Medium (DMEM)
Fetal Bovine Serum
Recommended Host:

Recommended Host: xa0BSC-40 (ATCC CRL-2761)

Production host: BSC-40 (ATCC CRL-2761) Alternate hosts: MRC-5, BSC-1, HeLa, LLC-MK2
Growth Conditions:
For best results, plate cells 24-48 hours in advance in DMEM with 10% FBS at 37.0°C. Infect at 90-100% confluence using a multiplicity of infection (MOI) of about 1. Infect and incubate in DMEM supplemented with 2% FBS at 32.0°C for 1-3 days. Harvest cells and culture fluids when 100% of culture shows advanced cytopathic effects. Vaccinia virus is largely cell associated. Concentrated preparations can be made by collection and sonication of cell pellets.
