Medium : 90% Leibovitzs L-15 medium + 10% h.i. FBS
Subculture : seed out at 2-4 x 106 cells/80 cm2; split confluent culture 1:2 to 1:6 using trypsin/EDTA; cells can partially be detached by tapping the flask; cells may need several days to re-adhere again
Incubation : at 37 °C, no exposure to CO2 (keep lid of flask closed)
Doubling time : ca. 50-60 hours
Harvest : about 25-35 x 106 cells/80 cm2
Storage : frozen with 85% medium, 10% FBS, 5% DMSO
Mycoplasma : contamination was eliminated with Ciprobay (ciprofloxacin), then negative in DAPI, microbiological culture, RNA hybridization, PCR assays

Fingerprint : multiplex PCR of minisatellite markers revealed a unique DNA profile
Cytogenetics : human hypotetraploid karyotype with 6% polyploidy and near diploid sideline (2%) - 89(85-91)<4n>XXX, -X - highly rearranged leading to extensive marker formation, including der(5)t(5;?14)(p11;q11)x2 - dm present in 12% cells
