Medium : 80% RPMI 1640 + 20% h.i. FBS + 10 µg/ml human insulin + 2 mM L-glutamine
Subculture : seed out at ca. 1-3 x 106 cells/25 cm2; split confluent culture 1:2 to 1:3 every 7-10 days using trypsin/EDTA; cells grow slowly and may produce cell debris
Incubation : at 37 °C with 5% CO2
Doubling time : ca. 100 hours
Harvest : about 1.0-1.6 x 105 cells/cm2 or 20 x 106 cells/175 cm2
Storage : frozen with 70% medium, 20% FBS, 10% DMSO
