Origin : originally described in the literature to be derived from the tumor-containing left axillary lymph node of a 39-year-old white man with small lung cell carcinoma (T3 N3 M3) in 1989; however, DNA fingerprinting and cytogenetic analysis showed cross-contamination with cell line RPMI-8226; RPMI-8226 was established from the peripheral blood of a 61-year-old man with multiple myeloma (IgG lambda-type) at diagnosis in 1966

Morphology: mostly adherent round cells growing in monolayers; image; image
Medium : 90% RPMI 1640 + 10% h.i. FBS
Subculture : seed out at ca. 2 x 106 cells/80 cm2; split confluent culture 1:4 to 1:6 every 3-4 days using trypsin/EDTA
Incubation : at 37 °C with 5% CO2
Doubling time : about 40 hours
Harvest : harvest of about 16 x 106 cells/80 cm2
Storage : frozen with 70% medium, 20% FBS, 10% DMSO
Mycoplasma : negative in DAPI, microbiological culture, RNA hybridization, PCR assays
