Morphology: single round cells growing in suspension, partly slightly adherent
Medium : 90% Dulbeccos MEM + 10% h.i. FBS + 4 mM L-glutamine
Subculture : split saturated culture optimally 1:2 to 1:3 every second day; seed out at ca. 0.5 x 106 cells/ml; maintain at ca. 0.3-0.4 x 106 cells/ml; maximal density at ca. 0.7 x 106 cells/ml; some cells attach to the flask surface, but may be detached by tapping the flask; if viability decreases, addition of HFCS ("Hybridoma Fusion and Cloning Supplement") may be useful
Incubation : at 37 °C with 5% CO2
Doubling time : ca. 24 hours
Harvest : cell harvest of ca. 0.5 x 106 cells/ml
Storage : frozen with 70% medium, 20% FBS, 10% DMSO
Mycoplasma : negative in DAPI, microbiological culture, RNA hybridization, PCR assays
Immunology : secretion of IgG2a monoclonal antibody
Cytogenetics : murine flat-moded hypertetraploid karyotype - 82-97<4n> - several centric fusion markers present
