Morphology: round to epitheloid cells growing as single cells or as clumps either in suspension or, if supplemented with EGF, in monolayers; image; image
Medium : 80-90% Dulbeccos MEM + 10-20% h.i. FBS + 2 mM L-glutamine + EGF (1 µg/100 ml, optional)
Subculture : seed out at ca. 0.2-0.5 x 106 cells/ml (suspension culture) or 0.4-0.8 x 105 cells/cm2 (adherent culture); maintain at about 0.2-1.5 x 106 cells/ml or equivalent; split confluent culture 1:4 to 1:8 every 3-5 days by agitation; cell number may be difficult to ascertain due to growth in clumps; cells grow faster in suspension without EGF, however EGF supports monolayer formation
Incubation : at 37 °C with 10% CO2
Doubling time : ca. 40 hours
Harvest : cell harvest of ca. 3 x 106 cells/ml or 1 x106 cells/cm2
Storage : frozen with 70% medium, 20% FBS, 10% DMSO
Mycoplasma : negative in microbiological culture, RNA hybridization, PCR assays

Fingerprint : multiplex PCR of minisatellite markers revealed a unique DNA profile
Cytogenetics : human near-diploid karyotype with 16% polyploidy - 45-48<2n>XX, +1, +7, -8, -13, +20, add(1)(p36), der(1;16)(q10;p10), del(2)(q11q2?2), del(13)(q1?1q1?3), der(16)t(1;16)(q12-21 - p11) - resembles originators unpublished karyotype
