Morphology: single, round to polymorph cells growing in suspension; image
Medium : 80-90% RPMI 1640 + 10-20% h.i. FBS
Subculture : seed out and maintain at ca. 0.5-1.0 x 106 cells/ml; it is recommended to start culture in a 24-well-plate and with 20% FBS; split saturated culture 1:2 to 1:4 every 2-3 days; viability of culture may drop to about 50% after thawing and may need more than one week to recover
Incubation : at 37 °C with 5% CO2
Doubling time : ca. 30-50 hours
Harvest : maximum density at about 1 x 106 cells/ml
Storage : frozen with 70% medium, 20% FBS, 10% DMSO
Mycoplasma : negative in microbiological culture, PCR assays
