Medium : 90% RPMI 1640 + 10-20% h.i. FBS
Subculture : seed out at ca. 0.8 x 106 cells/ml initially in a 24-well-plate; viability may drop below 50% during the first few days and a significant amount of cell debris is always visible; in logarithmic growth phase split culture 1:2 to 1:3 every 2-3 days and maintain at ca. 0.5-2.0 x 106 cells/ml; maximal density at about 3 x 106 cells/ml
Incubation : at 37 °C with 5% CO2
