Morphology: adherent fibroblastoid cells growing as monolayer; image; image; image
Medium : 90% Dulbecco´s MEM + 10% h.i. FBS + (additionally) 2 mM L-glutamine (to sustain genetic stability cells should be maintained, at least periodically, with 0.5 µg/ml puromycin)
Subculture : culture vessels should be coated with 0.01% poly-L-lysine solution; seed out at 1 x 106 cells/25 cm2; split subconfluent culture 1:3 to 1:4 every 2-3 days using trypsin/EDTA; optimal density around 4 x 106 cells/25 cm2
Incubation : at 37 °C with 10% CO2
Doubling time : ca. 24 hours
Harvest : cell harvest of ca. 8 x 106 cells/25 cm2 flask
Storage : frozen with 70% medium, 20% FBS, 10% DMSO
