A sister cell line with the retrovirally reconstituted XRCC2 gene is available as ACC 853

Morphology: adherent fibroblastoid cells growing as monolayer with formation of islets; image; image
Medium : 90% Dulbecco´s MEM + 10% h.i. FBS + (additionally) 2 mM L-glutamine (to sustain genetic stability cells should be maintained, at least periodically, with 0.5 µg/ml puromycin)
Subculture : culture vessels should be coated with 0.01% poly-L-lysine solution; seed out at 1 x 106 cells/25 cm2; split 1:2 to 1:4 every 2-4 days using trypsin/EDTA; please note, cells grow really slow, when seeded out with lower density
Incubation : at 37 °C with 5% CO2
Doubling time : ca. 50-70 hours
Harvest : cell harvest of ca. 20 x 106 cells/80 cm2 flask
Storage : frozen with 70% medium, 20% FBS, 10% DMSO
